The Synaptril Formula: What the 575 mg Math Shows

Quick answer

The Synaptril formula is a 575 mg proprietary blend of six ingredients, and 80 mg of its magnesium comes from magnesium glycinate. Simple label math says that compound likely takes 400 to 570 mg, leaving about 5 to 175 mg for the other five. These are estimates, not lab results.

The formula01

What is the Synaptril formula?

The Synaptril formula is one proprietary blend of 575 mg per capsule, made of six ingredients. The label prints the total and the order, but not the share of each line.

It also prints one extra number: 80 mg of magnesium, 19% of the Daily Value, "as Magnesium Glycinate from proprietary blend". That number is the key to the math, because it ties one blend line to a known amount.

No other line has a printed amount. So a reader must work from the weights that the numbers allow, as the next sections do (21 CFR 101.36, supplement labeling).

Think of the capsule as a small box with a fixed weight. One item in the box has a known size. The label lists the other five in order, so each one must be smaller than the one before it.

Three steps02

How much of the blend is magnesium glycinate?

Magnesium glycinate likely makes up 400 to 570 mg of the 575 mg blend. That follows from one fact: the compound is only about 14% magnesium by formula weight, and buffered grades reach about 20%.

Divide 80 mg by 0.20 and you get 400 mg. Divide 80 mg by 0.14 and you get about 570 mg. Whatever the grade, the first line takes most of the blend.

Subtract that from 575 mg and little remains. At 400 mg of glycinate, 175 mg is left. At 570 mg, only 5 mg is left. The truth lies somewhere between, and the label does not say where.

  5. 5Turmeric (95% curcuminoids)not printed

Step 1. The label prints 80 mg of magnesium "as magnesium glycinate from proprietary blend".

Step 2. By formula weight, magnesium glycinate is about 14% magnesium; buffered grades run up to about 20%. So 80 mg of magnesium needs about 400-570 mg of the compound.

Step 3. 575 mg minus that leaves about 5-175 mg for alpha-lipoic acid, butcher's broom, L-carnitine, turmeric and CoQ10 together.

Our estimate from the label, not a lab test. A blend has to list its total and its order, not single amounts (21 CFR 101.36, supplement labeling). Magnesium figures from NIH (NIH ODS, Magnesium).

Our estimate from the label, not a number the maker printed.

What remains03

How much is left for the other five ingredients?

About 5 to 175 mg is left for alpha-lipoic acid, butcher's broom, L-carnitine, turmeric and CoQ10 together. Shared five ways, each line is small. If the five were equal, each would hold between 1 and 35 mg.

The label order adds a second limit. Line k weighs at most 575 divided by k, and after the glycinate it can weigh at most 175 divided by k minus one.

Most each line can weigh, from the label order and the glycinate math
LineOrder ceilingTighter estimateStudied dose
Alpha-lipoic acid288 mgunder about 175 mg600 mg
Butcher's broom192 mgabout 88 mgNo brain dose
L-carnitine144 mgabout 58 mg1.5 to 3 g (acetyl form)
Turmeric extract115 mgabout 44 mg90 mg curcumin twice daily
Coenzyme Q1096 mgabout 35 mg30 to 100 mg typical

For L-carnitine the studied dose is of a different form, so the gap is wider than it looks (NIH ODS, Carnitine).

What it means04

What does the formula math mean for you?

It means the capsule is mainly a magnesium product, with small and unknown amounts of five other things. That is not bad. It is just not what the front claims suggest.

It also means the studies on the other five ingredients do not map onto this capsule. Studies of alpha-lipoic acid used 600 mg a day, and our tighter ceiling sits well under that (Linus Pauling Institute, Lipoic Acid).

The fair way to treat the formula is as a gentle magnesium capsule with a long ingredient list. Ask whether you want that, and at that price.

At $49 a bottle on the six-bottle pack, a capsule costs about $1.63 a day. At $89 a bottle, it costs about $2.97. Weigh those against what a plain magnesium product would cost.

A maker could settle the question by printing every amount. Until then, the math is the best we can do. Read the full label on our supplement facts page, and try the ingredients page for each line.

FAQ05

Questions about the formula math

Why does the label math use 14% and 20%?

Magnesium glycinate is about 14% magnesium by formula weight. Buffered grades with extra magnesium can reach about 20%. The two figures give the low and high ends of our range.

Can the blend math prove the exact amounts?

No. It gives limits, not exact amounts. Only the maker can print the true split, or a lab test can find it. We say likely, about and at most for that reason.

Is a smaller dose of an ingredient harmful?

Not by itself. A small amount is less likely to cause side effects, but it is also less likely to match any studied dose. The label cannot tell you which side you are on.
